I'd be satisfied if the game turns out at least as good as the game that trailer's for. Only on Planet U (the trailer is set on a mythological earth); futuristic science (trailer is steampunk); energy crystals, rock monsters, and all the RR goodies; multiple Z levels (You've implied RR2 will have multiple Z levels, which makes me very excited; and less eldritch horrors (which the trailer has -- just replace with more RR goodies ).
Multiplayer is also a big feature for me.
I personally want a realistic graphics style.
If you add a health system similar to Dwarf Fortress (or a little less complicated -- as in GURPS), I'd be very excited.
Optionally, you could make it a "Losing is FUN!" game. Because, really, it is. Winning isn't "Level complete!" That's boring, and can cut off the fun of playing. The best fun comes from the narratives the game produces, though a player necessarily have to "lose" the game to achieve that -- it's an ongoing process of continuously playing the game and restarting when you lose.
